Sorry for the irregular updates. Not able to update on a
regular basis. It was almost 12 in the night by the time I reached room
yesterday. The days go un noticed with almost 6-8 PCR cycles per day, running
them on gels and in analyzing their results. The attempts sometimes give poor
results and they must be repeated for quite several times. Apart from screening
primers in the parents and identifying them in mapping population, I get a
chance to learn about running SDS- PAGE gels on which one of our team mates is
carrying on her experiments of studying proteins present in starch isolated
from wheat. In the process, I learnt how
to isolate proteins from starch samples for SDS -PAGE.
Today, observed the work being done on western blotting. The
proteins were immobilized on PVDF Membrane. Then, with the use of primary,
secondary antibodies and substrate, color development was detected colorimetrically.
It gives a great feeling to relate the theory learnt
regarding these techniques and to observe them practically. Science is more interesting
when done practically than in books.
It s third day I've spoken to my seeds as a part of the experiment to know whether there are any changes in plants based on how humans treat them. Eager to know how it will be working.Signing Off. Good Night.
