The atmosphere here at Mohali has changed since two days. It has become dusty and it seems like dust particles have been trapped and are suspended in the air. Badly waiting for a shower that can clean up the atmosphere. Even the night temperatures are high making it a little uncomfortable.
Coming back to work, today learnt couple of new things at the lab. Observed RNA isolation procedure. Since, RNA is very unstable and being single stranded, it is too fragile and extra care is taken while isolating it. It is usually done in a special cabin called the RNA Room. Gloves are a must while handling RNA because our skin produces specific RNases that usually are antipathogenic and protect us from pathogens and UV radiations. The chemicals used during the isolation process are known to cause skin burns and scarring.
The isolated RNA was examined on 0.8% agarose gel and quantified on NanoDrop. Then proceeded for cDNA synthesis. It was the first time to visualize cDNA synthesis process practically. My senior explained it's protocol and steps involved in it. The synthesized DNA was examined by PCR amplification. It was followed again by agarose gel electrophoresis to check whether the cDNA was synthesized. It gave positive results, so proceeded for amplifying the specific gene from the cDNA using primers specific for the gene of interest. So,the PCR is in process now. It will take about 1 and a half hour more.
