Restriction Digestion: It is the process of cutting DNA molecules into smaller pieces by using enzymes called restriction enzyme which identify specific restriction sites on DNA.
Elution of DNA: After running the DNA on gel electrophoresis, often it is necessary to identify the required band of DNA and use it for further analysis or processing. The process of obtaining or separating the DNA from the Agarose gel is called Elution.
Ligation: It is the joining of DNA strands or nicks in DNA by forming phosphate ester bond.
Principle for Elution of DNA from the Gel: (brief)
The portion of gel containing the desired band of DNA is excised by using a scalpel blade. The gel containing DNA is dissolved in QG Buffer and then treated with isopropanol, PE buffer and washed with Ethanol. After all the treatments, DNA is obtained free of Agarose, buffers and other chemicals.
