Day 17: All in Vain

Today's task was about estimating amylose from approximately 50 starch samples of wheat. 
1. Plotting standard curve 
2. Estimating amylose content from starch samples.

- starch sample does not dissolve in water. So, DMSO (Dimethyl Sulphonate) is used as solvent
- Then the samples are allowed to rest for 10 minutes in boiling water bath at 85C
- Amylose content is estimated by using Spectrophotometry
- Iodine is used in the process for estimation because Starch- Iodine complex gives a blue-violet coloration which can be detected using spectrophotometer at 620 nanometer.

A very long and tiring day from 9:00 am to 10:30 pm. But at the end, the results weren't satisfactory. Spectrophotometer probably got too tired and collapsed because it was continually under use for past two days. Hoping for good start tomorrow.
