Today, made progress in learning basic techniques.
Learnt how to elute DNA from the Gel with detailed procedure,for using the DNA bands for restriction digestion or ligation.
My project on use of SSR Markers in construction of Linkage maps marked it's start. Tested the DNA isolated from two diverse Parents of wheat(P1 and P2) using 35 SSR primers by performing PCR.
Out of 35 Primers, only 3 primers showed polymorphism. More primers are to be tested.
Techniques learnt today- demonstration by guide:
1. DNA Elution
2. Preparing 5x TBE buffer and its dilutions
3. Making PCR reaction mixture
4. Setting up PCR
5. Gel Electrophoresis
6. Reading the Gel through documentation system and identifying polymorphism.
